MOTS-c Peptide Overview
MOTS-c (mitochondrial open-reading-frame of the 12S rRNA-c) is a novel mitochondria-derived peptide composed of 16 amino acids. It is expressed in tissues and plasma, functioning as both a cell-specific compound and a “mitokine” or mitochondrial hormone. Research suggests it may translocate to cellular nuclei during metabolic stress to alter gene expression, primarily by stimulating the AMP-activated protein kinase (AMPK) pathway.

Research and Clinical Studies
- Metabolism & Aging: Studies indicate MOTS-c levels may decline with age, contributing to age-related metabolic dysfunction. It may interact with aging regulators like NAD+ and sirtuins to modulate cell lifespan.
- Muscle Metabolism: Research suggests the peptide may improve insulin sensitivity in skeletal muscles by enhancing glucose transporter expression via AMPK activation. It may also target the folate-methionine cycle and purine biosynthesis to modulate nutrient sensing.
- Fat Cell Metabolism: In murine models on high-fat diets, MOTS-c appeared to prevent fat accumulation and induce glucose uptake. It may regulate stress-responsive transcription factors like NRF2 to improve metabolic homeostasis and reduce diet-induced obesity.
- Bone Health: The peptide has been suggested to promote osteogenic differentiation of bone marrow mesenchymal stem cells via the TGF-β/Smad pathway, potentially improving bone density and strength.
- Cardiac Function: While not directly affecting the heart, researchers suggest MOTS-c may improve the function of endothelial cells lining blood vessels, which could help regulate blood pressure and clotting.
- Cell Lifespan: Some research suggests a biological link between MOTS-c and extended longevity, though further mechanistic research is needed to understand how specific structural polymorphisms affect its endocrine action.
