NAD+ Peptide Overview
Nicotinamide Adenine Dinucleotide (NAD+) is an endogenous nucleotide that regulates primary functions such as metabolism, energy production, and DNA repair. It acts as a secondary messenger and is synthesized naturally via the de novo mechanism from the amino acid tryptophan. Research suggests it exerts over 500 enzymatic reactions and acts as a coenzyme in redox functions.
Mechanism of Action
NAD+ is a coenzyme for three major classes of enzymes:
- Sirtuins (SIRTs): May stimulate mitochondrial homeostasis, stem cell regeneration, and nerve protection.
- Poly ADP ribose polymerases (PARPs): Synthesize polymers that lead to genome stability.
- Cyclic ADP ribose synthetases (cADPRS): Key immunological cells (CD38 and CD157) that may stimulate stem cell regeneration and DNA repair.
Chemical Makeup

Research and Clinical Studies
- Productive Aging: Studies in mice suggest that NAD+ intermediates (NMN and NR) may reduce weight gain, increase energy metabolism, and improve lipid profiles.
- Neurodegenerative Activity: Research on aged mice models suggests mitochondrial functions can be restored in nerve and brain cells when exposed to NAD+ intermediates.
- DNA Repair: Studies indicate that replenishing NAD+ can significantly improve DNA base excision repair activity and cell viability after ischemic stress (oxygen/glucose deprivation).
- Organ Health (Liver & Kidney): Experimental models suggest NAD+ may help prevent obesity and alcoholic hepatitis while protecting against glucose-induced kidney cell hypertrophy.
- Skeletal & Cardiac Function: Research indicates NAD+ plays a vital role in cellular respiration (glycolysis and the citric acid cycle). In cardiac studies, it reportedly produced cardioprotective functions against ischemic injury in mice.
